The Vasco da Gama made its maiden voyage to Gqeberha having previously stopped at ports in Cape Town and Mossel Bay.
The majestic cruiseliner's presence is seen as a major boost to the economy.
At least 28 cruise ships are expected to descend on Gqeberha during the festive season.
Transnet's National Ports Authority says it will ensure its visitors safety and comfort through sufficient marine services, including berth availability and accommodation of crews.
